Özet:

The In Vitro Effect of Benzene on Liver Glutathione S-transferase Enzyme Activity" In this study, the effect of benzene which is known as a carcinogen and toxic agent on liver Glutathione S-transferase (GST) enzyme was studied in vitro. GST enzyme activity was determined by following the formation of l-chloro-2,4- dinitrobenzene -Glutathione (CDNB-GSH) conjugate with spectroscopic method. Our results showed that there is an inverse correlation between GST conjugation activity and concentration of benzene, and reaction time. Variations in the Vmax and Km value for CDNB of the enzyme was observed. The results obtained showed that GST activity for CDNB-GSH conjugation reaction was inhibited with mixed reaction mechanism by benzene. Key Words : Benzene, Glutathione, Glutathione S-transferase
