Flavonoid ailesinin bir üyesi olan kalkonlar, antikanser etkiye sahip olduğu bilinen doğal bileşiklerdir. Kalkon ve sentetik türevleri, günümüzde kanser çalışmalarda yoğun olarak araştırılmaya devam etmektedir. Bu çalışmada, Claisen-Schmidt reaksiyonu ile sentezlenen yeni bir Kalkon türevi bileşiğin 1-(7-etoksi-1-benzofuran-2-il)-5-(4-hidroksi-3-metoksifenil)penta-2,4-dien-1-on insan akciğer (A549, H1299) ve kolon kanseri (HCT116, HT29) hücrelerinde antikanser aktivitesinin araştırılması amaçlandı. Kalkon bileşiğinin hücre canlılığına etkisi SRB testi ile değerlendirildi ve kemoterapi ilacı olarak kullanılan 5-FU ile kombinasyon çalışması yapıldı. Hücre ölümü modu, Hoechst 33342, Annexin-V-FITC ve Propidium iodide (PI) üçlü boyama ile floresan görüntüleme yöntemiyle belirlendi. Kalkon bileşiğinin IC50 değerleri A549, H1299, HCT116, HT29 için sırasıyla 2.85, 1.46, 0.59, 0.35 uM olarak bulundu. Floresan görüntüleme sonucunda, Annexin-V-FITC (yeşil) ile pozitif boyamaya ek olarak hücrelerde piknotik çekirdekler ve kromatin yoğunlaşması gözlendi. Sonuçlar, yeni sentezlenen Kalkon türevi bileşiğin kanser hücreleri üzerinde önemli bir sitotoksik etkiye sahip olduğunu ve apoptozu indüklediğini gösterdi. A member of the flavonoid family, chalcones are natural compounds known to have anticancer effects. Chalcone and its synthetic derivatives continue to be intensively investigated in cancer studies today. In this study, it was aimed to investigate the anticancer activity of a new Chalcone derivative compound (1-(7-etoksi-1-benzofuran-2-il)-5-(4-hidroksi-3-metoksifenil)penta-2,4-dien-1-on), synthesized by the Claisen-Schmidt reaction, in human lung (A549, H1299) and colon cancer (HCT116, HT29) cells. The effect of chalcone compound on cell viability was evaluated with the SRB test and a combination study was performed with 5-FU, which is used as a chemotherapy drug. Cell death mode was determined by fluorescence imaging with Hoechst 33342, Annexin-V-FITC and Propidium iodide (PI) triple staining. The IC50 values of chalcone compound were found as 2.85, 1.46, 0.59, 0.35 μM for A549, H1299, HCT116, HT29, respectively. As a result of fluorescence imaging, pycnotic nuclei and chromatin condensation were observed in the cells in addition to positive staining with Annexin-V-FITC (green). The results showed that the newly synthesized Chalcone derivative compound had a significant cytotoxic effect on cancer cells and induced apoptosis.
