Cynarin and oleuropein in type 1 diabetic rats: Effects on the oxidative and antioxidative systems

Oxidative stress can be induced by blood glucose and lipid levels in Diabetes Mellitus, which can lead to the emergence of diabetes complications. However, cynarin and oleuropein can decrease oxidative stres by reducing enhanced-blood glucose and lipid levels, and effecting antioxidant enzyme systems. This study was designed to investigate the effects of oleuropein and cynarin on hypoglycemic, oxidant and antioxidant systems in streptozotocin induced type 1 diabetes. Type 1 diabetes were constituted with injection intraperitoneally one dose of streptozotocin (65mg/kg). Cynarin, oleuropein and cynarin+oleuropein (%10) was supplemented in drinking water for 5 weeks. Wistar rats were randomly divided into eight groups; control (C), control+oleuropein (C+OLEE), control+cynarin (C+CYE), control+oleuropein+cynarin (C+OLEE+CYE), diabet (D), diabet+oleuropein (D+OLEE), diabet+cynarin (D+CYE) and diabet+oleuropein+cynarin (D+OLEE+CYE). Serum triglyceride, plasma and tissue MDA levels were observed to be significantly reduced while serum total antioxidant capacity were significantly increased in control oleuropein, control cynarin and control oleuropein+cynarin group when compared with control group. Tissue MDA, blood glucose, serum total cholesterol and triglyceride levels were signigicantly reduced while serum insulin, blood glutathione peroxidase and erythrocyte superoxide dismutase, serum paraoxonase and arylesterase activities were significantly increased in diabet oleuropein, diabet cynarin and diabet oleuropein+cynarin group when compared with diabet group. In conclusion, with this study, it reported that oleuropein and cynarin have antihyperglycemic, antihyperlipidemic and antioxidant features, and besides that they have a protective and preventive effect against oxidative stres. It can be used to supplement and support the treatment of diabetes on rats which were exposed to type 1 diabetes.
